    High throughput genomic sequencing of bioaerosols in broiler chicken production facilities

    Chronic inhalation exposure to agricultural dust promotes the development of chronic respiratory diseases among poultry workers. Poultry dust is composed of dander, chicken feed, litter bedding and microbes. However, the microbial composition and abundance has not been fully elucidated. Genomic DNA was extracted from settled dust and personal inhalable dust collected while performing litter sampling or mortality collection tasks. DNA libraries were sequenced using a paired-end sequencing-by-synthesis approach on an Illumina HiSeq 2500. Sequencing data showed that poultry dust is predominantly composed of bacteria (64–67%) with a small quantity of avian, human and feed DNA (\u3c 2% of total reads). Staphylococcus sp. AL1, Salinicoccus carnicancri and Lactobacillus crispatus were the most abundant bacterial species in personal exposure samples of inhalable dust. Settled dust had a moderate relative abundance of these species as well as Staphylococcus lentus and Lactobacillus salivarius. There was a statistical difference between the microbial composition of aerosolized and settled dust. Unlike settled dust composition, aerosolized dust composition had little variance between samples. These data provide an extensive analysis of the microbial composition and relative abundance in personal inhalable poultry dust and settled poultry dust

    The marijuana legalization trend promotes the notion that cannabinoids such as tetrahydrocannabinol (THC) can be safe and effective medications for conditions such as insomnia. Unfortunately, exposure to THC during development has been shown to impair cognition and motor behavior in humans and laboratory animals. The current study used VitalView software and cage-top infrared activity monitors to quantify the effects of a range of THC doses on activity and circadian rhythms in adult rats that were exposed to THC throughout adolescence. Rats were further administered acute nicotine drug challenges to determine if THC altered the development of the acetylcholine neurotransmitter system. It is hoped that the results of this work in progress can be used to determine if THC presents a risk to adolescent brain development and predisposes an individual to future activity and sleep dysregulation

    The Intersection of Gender, Media, And Policy: A Qualitative Analysis on Thai Newspaper Coverage of Women in Agriculture

    Women empowerment and gender equality have been found to be statistically significant and positive predictors of global agricultural development. Therefore, reducing gender disparities can encourage economic progress and growth in developing nations. As such, determining effective ways to stimulate social progress and women’s empowerment has emerged as a critical need. One strategy used to raise the public’s consciousness about gendered issues in Thailand has been through mass media. In response, this study aimed to (1) determine to what extent Thailand’s newspaper coverage focused on topics related to women and the agricultural industry; and (2) describe how women in agriculture have been portrayed in newspaper coverage since the introduction of Thailand 4.0’s policy in 2016. Using a qualitative content analysis of Thailand’s newspaper coverage of women in the agricultural sector, four themes emerged: (1) economic policy implications for Thailand’s agricultural system; (2) human rights; (3) women entrepreneurship and leadership; and (4) agricultural development. Therefore, this study concluded that newspaper coverage of women in agriculture was diverse and conflicting – a finding not previously reported. The findings also revealed that women in agriculture have been underrepresented in newspaper publications historically. Moving forward, we provided critical implications for how future research, theory, and practice can depict women in agriculture more positively in the newspaper media

    Casting a Critical Lens on Thailand’s Higher Education System: A Case Study of Women’s Experiences as Agricultural Extension Faculty

    The agricultural industry and higher education have traditionally been male-dominated spaces in the developing world. However, in recent decades, significant progress in female representation has been achieved in both sectors. Previous research has suggested that women in the Southeast Asian agricultural industry have been more empowered than women in other regions. However, women in Thailand’s agricultural postsecondary programs have been understudied. In response, this study examined the experiences and perceptions of women agricultural extension faculty in Thailand’s higher education system. Through qualitative analysis of in-depth interviews with women, three distinct themes emerged: (1) gendered disparities, (2) barriers to success in academia, (3) perceptions of self and gender in agriculture and higher education. The findings suggested that Thai female faculty in agricultural extension held positive perceptions of their careers despite persistent gender inequalities and traditional cultural norms. Moving forward, we recommend that more in-depth research be conducted to understand better how such perceptions coexist with the deeply entrenched gender inequalities

    Occurrence of Potentially Zoonotic and Cephalosporin Resistant Enteric Bacteria among Shelter Dogs in the Central and South-Central Appalachia

    BACKGROUND: Antimicrobial resistance and presence of zoonotic enteropathogens in shelter dogs pose a public health risk to shelter workers and potential adopters alike. In this study we investigated the prevalence of zoonotic bacterial pathogens and cephalosporin resistant (CefR) enteric bacteria in the feces of apparently healthy shelter dogs in the Cumberland Gap Region (CGR) in the US states of Kentucky, Tennessee and Virginia. RESULTS: Fecal samples of 59 dogs from 10 shelters in the CGR of Central and South-Central Appalachia were screened for the presence of Campylobacter jejuni, Clostridium perfringens, Salmonella and CefR enteric bacteria. C. jejuni, C. perfringens were detected by PCR based assays. Culture and PCR were used for Salmonella detection. Of 59 dogs, fecal samples from 14 (23.7%) and 8 (13.6%) dogs tested positive for cpa and hipO genes of C. perfringens and C. jejuni, respectively. Salmonella was not detected in any of the tested samples by PCR or culture. CefR enteric bacteria were isolated on MacConkey agar supplemented with ceftiofur followed by identification using MALDI-TOF. Fecal samples from 16 dogs (27.1%) yielded a total of 18 CefR enteric bacteria. Majority of CefR isolates (14/18, 77.8%) were E. coli followed by, one isolate each of Enterococcus hirae, Acinetobacter baumannii, Acinetobacter pittii, and Pseudomonas aeruginosa. CefR enteric bacteria were tested for resistance against 19- or 24-antibiotic panels using broth microdilution method. Seventeen (94.4%) CefR bacteria were resistant to more than one antimicrobial agent, and 14 (77.8%) displayed multidrug resistance (MDR). CONCLUSIONS: This study shows that shelter dogs within the CGR not only carry zoonotic bacterial pathogens, but also shed multidrug resistant enteric bacteria in their feces that may pose public health risks

    The Long-Baseline Neutrino Experiment: Exploring Fundamental Symmetries of the Universe

    The preponderance of matter over antimatter in the early Universe, the dynamics of the supernova bursts that produced the heavy elements necessary for life and whether protons eventually decay --- these mysteries at the forefront of particle physics and astrophysics are key to understanding the early evolution of our Universe, its current state and its eventual fate. The Long-Baseline Neutrino Experiment (LBNE) represents an extensively developed plan for a world-class experiment dedicated to addressing these questions. LBNE is conceived around three central components: (1) a new, high-intensity neutrino source generated from a megawatt-class proton accelerator at Fermi National Accelerator Laboratory, (2) a near neutrino detector just downstream of the source, and (3) a massive liquid argon time-projection chamber deployed as a far detector deep underground at the Sanford Underground Research Facility. This facility, located at the site of the former Homestake Mine in Lead, South Dakota, is approximately 1,300 km from the neutrino source at Fermilab -- a distance (baseline) that delivers optimal sensitivity to neutrino charge-parity symmetry violation and mass ordering effects. This ambitious yet cost-effective design incorporates scalability and flexibility and can accommodate a variety of upgrades and contributions. With its exceptional combination of experimental configuration, technical capabilities, and potential for transformative discoveries, LBNE promises to be a vital facility for the field of particle physics worldwide, providing physicists from around the globe with opportunities to collaborate in a twenty to thirty year program of exciting science. In this document we provide a comprehensive overview of LBNE's scientific objectives, its place in the landscape of neutrino physics worldwide, the technologies it will incorporate and the capabilities it will possess.Comment: Major update of previous version.     Phenome-wide association analysis of LDL-cholesterol lowering genetic variants in PCSK9

    Abstract: Background: We characterised the phenotypic consequence of genetic variation at the PCSK9 locus and compared findings with recent trials of pharmacological inhibitors of PCSK9. Methods: Published and individual participant level data (300,000+ participants) were combined to construct a weighted PCSK9 gene-centric score (GS). Seventeen randomized placebo controlled PCSK9 inhibitor trials were included, providing data on 79,578 participants. Results were scaled to a one mmol/L lower LDL-C concentration. Results: The PCSK9 GS (comprising 4 SNPs) associations with plasma lipid and apolipoprotein levels were consistent in direction with treatment effects. The GS odds ratio (OR) for myocardial infarction (MI) was 0.53 (95% CI 0.42; 0.68), compared to a PCSK9 inhibitor effect of 0.90 (95% CI 0.86; 0.93). For ischemic stroke ORs were 0.84 (95% CI 0.57; 1.22) for the GS, compared to 0.85 (95% CI 0.78; 0.93) in the drug trials. ORs with type 2 diabetes mellitus (T2DM) were 1.29 (95% CI 1.11; 1.50) for the GS, as compared to 1.00 (95% CI 0.96; 1.04) for incident T2DM in PCSK9 inhibitor trials. No genetic associations were observed for cancer, heart failure, atrial fibrillation, chronic obstructive pulmonary disease, or Alzheimer’s disease – outcomes for which large-scale trial data were unavailable. Conclusions: Genetic variation at the PCSK9 locus recapitulates the effects of therapeutic inhibition of PCSK9 on major blood lipid fractions and MI. While indicating an increased risk of T2DM, no other possible safety concerns were shown; although precision was moderate
